Understanding the role and behaviour of structural proteins within their biological environment is a difficult challenge especially as they generally have hierarchical structures on many length scales which give rise to their properties. The major mammalian structural protein is keratin and is present in hair and skin. The hair cortex is responsible for the majority of the strength of a hair as well as its colour; it consists of coiled-coils of keratin, known as intermediate filaments (IF’s) ~90A in diameter embedded in a matrix of mainly Keratin Associated Proteins (KAP’s).

Fischer-Tropsch (FT) synthesis can support the transition to a net zero economy by converting municipal waste into sustainable aviation fuels (SAFs). Co-based catalysts have proven highly efficient for FT synthesis, and an understanding of these catalyst has been developed over several years with promotional additives critical during synthesis, catalyst treatment, and FT synthesis [1,2]; however, how these promoters affect the chemistry remains poorly understood.

Summary of PhD Project
This project will capitalise on recent developments human Trophoblast (TSC) and induced pluripotent stem cell (iPSC) technologies including 3D placental and embryo organoid culture, together with CRISPR, EPI-CRISPR to determine how epigenetic mechanisms influence development and the production of placental hormones. A master regulator of placental endocrine lineage development will be targeted genetically (CRISPR) and epigenetically (EPI-CRISPR) in male and female human TSC lines which will then be differentiated in organoids to access their capacity to produce endocrine cells and secrete placental hormones.

Adult neural stem cells (aNSCs) in the mammalian brain can both self-renew and generate new neurons throughout life in a process called adult neurogenesis. Understanding the molecular mechanisms that regulate aNSCs is critical because adult neurogenesis is necessary for learning, mood control, and body weight maintenance. Our recently published results show that the transcription factor ZEB1 is essential for the self-renewal of aNSCs and continuous neurogenesis in the hippocampus, a brain area involved in learning and emotion.

This relatively small population (O(1)% of the total) will have a tremendous impact on our understanding of high-mass stellar astrophysics. With future gravitational-wave detectors, we will observe hundreds of mergers that previously went through an X-ray binary phase.

This PhD is part of a larger EPSRC funded project to develop new catalysts for acetylene hydrogenation and hydrochlorination. Catalysts of gold and platinum will be synthesized, with nitrogen-containing ligands added to try to improve the selectivity and activity of catalysts. The PhD researcher on this project will determine the structure of the catalysts using atomic-resolution electron microscopy, with the ability to image individual metal atoms. The determined catalyst structure will allow the team to relate how catalyst structure affects catalytic performance, including direct use of the atomic structures in computational chemistry simulations.
